Forensic Context

A study in humans demonstrated that circALAS2 is used for body fluid identification, specifically for detecting menstrual blood, where detecting circRNAs derived from the same gene as mRNA typing could improve the stability and sensitivity of this identification [Li et al. DOI:10.1093/fsr/owad003]. A study in human body fluid samples demonstrated that the inclusion of circular RNA transcripts of the circALAS2 in mRNA profiling significantly improved detection sensitivity and stability for peripheral blood identification [Zhang et al. DOI:10.1007/s00414-017-1690-7]. The use of primers targeting both linear and circular forms enabled detection down to 0.003 ng of input RNA and provided robust detection in environmentally challenged, degraded, and aged (6-7 years) bloodstains where primers targeting only linear transcripts failed. A subsequent characterization confirmed the circALAS2 as a blood-specific biomarker and found that the inclusion of its circRNAs did not impair specificity [Liu et al. DOI:10.1007/s00414-019-02027-y].
