Forensic Context

A study in mice demonstrated that circNUBPL is highly expressed in the brain and cerebellum, where it regulates neuronal plasticity and structural changes in the synapse during development [Sema Misir et al. DOI:10.1038/s41418-022-00948-7]. A study in humans demonstrated that the circNUBPL is a highly specific circular RNA marker for semen identification, as part of a target RNA sequencing panel designed for body fluid contributor deconvolution in forensic mixtures [Yu et al. DOI:10.1016/j.fsigen.2025.103416]. The assay, which included 135 RNA-SNPs across 42 genes, showed that the circNUBPL exhibited high specificity for semen with no reported cross-reactivity in the tested samples, contributing to the panel's successful detection of all body fluid components in artificial mixtures and its high cumulative discriminatory power.
