Forensic Context

A study in human forensic samples demonstrated that the mir-219a was selected as the best performing marker for brain tissue identification based on pronounced separation properties (ΔΔCq >9.5) in pooled samples, which was confirmed in individual native specimens and dried swabs [Sauer et al. DOI:10.1016/j.fsigen.2017.02.002]. The validation using Binary Logistic Regression showed robust identification, achieving 100% correct classification for most native and dry brain samples, and the marker performed well in aged dry swabs (28 days), mixtures where no false positives occurred, and mock case samples from experimental shootings and stabbings, where brain was correctly identified even as a minor component in trace amounts. A study in mice demonstrated that the mir-219a targets CACUL1, a gene involved in caspase-3-dependent apoptosis regulated by Akt/Foxo3a and p53/Bcl-2 pathways [Matyasova et al. DOI:10.4149/gpb_2021038]. In human studies, the mir-219a is elevated in the serum of TBI patients, identifying it as a promising diagnostic biomarker [Li et al. DOI:10.1016/j.neuint.2023.105643].
