Summary

This gene encodes one of two cannabinoid receptors. The cannabinoids, principally delta-9-tetrahydrocannabinol and synthetic analogs, are psychoactive ingredients of marijuana. The cannabinoid receptors are members of the guanine-nucleotide-binding protein (G-protein) coupled receptor family, which inhibit adenylate cyclase activity in a dose-dependent, stereoselective and pertussis toxin-sensitive manner. The two receptors have been found to be involved in the cannabinoid-induced CNS effects (including alterations in mood and cognition) experienced by users of marijuana. A study in human fetal and adult tissues demonstrated that the CB1 cannabinoid receptor protein and mRNA are identifiable via immunohistochemistry and RT-PCR in long-term formalin-fixed paraffin-embedded samples, confirming the usability of such degraded tissues for molecular analysis [Blanchot et al. The experimental approach successfully detected the CNR1 in fetal brain regions and placenta, with differences in RNA extraction and PCR success observed across brain regions and fixation durations, establishing a methodological prerequisite for investigating cannabis toxicity during pregnancy.
