This gene encodes the folylpolyglutamate synthetase enzyme. This enzyme has a central role in establishing and maintaining both cytosolic and mitochondrial folylpolyglutamate concentrations and, therefore, is essential for folate homeostasis and the survival of proliferating cells. This enzyme catalyzes the ATP-dependent addition of glutamate moieties to folate and folate derivatives. Alternative splicing results in transcript variants encoding different isoforms. A study in humans investigated the FPGS as a reference gene for normalizing quantitative PCR data in the context of gamma-hydroxybutyric acid intake, where it was identified via geNorm, NormFinder, and BestKeeper algorithms as one of the two most suitable normalizers alongside PPIB [Mehling et al. In a separate forensic study for age estimation from blood, the FPGS was incorporated as one of nine reference genes in a targeted massively parallel sequencing assay, where it demonstrated stable expression and was included in the final validated primer pool [Hänggi et al.
