This gene encodes a protein which belongs to the C2H2-type zinc finger protein subclass of the Gli family. Members of this subclass are characterized as transcription factors which bind DNA through zinc finger motifs. These motifs contain conserved H-C links. Gli family zinc finger proteins are mediators of Sonic hedgehog (Shh) signaling and they are implicated as potent oncogenes in the embryonal carcinoma cell. The protein encoded by this gene localizes to the cytoplasm and activates patched Drosophila homolog (PTCH) gene expression. It is also thought to play a role during embryogenesis. The encoded protein is associated with several phenotypes- Greig cephalopolysyndactyly syndrome, Pallister-Hall syndrome, preaxial polydactyly type IV, postaxial polydactyly types A1 and B. Forensic Context
A study in porcine skin demonstrated that cutaneous exposure to bromine vapor for 10 or 20 minutes significantly decreased the GLI2 transcript by approximately 3.0-fold at 48 hours post-exposure, identifying it as a component within altered signaling pathways [Rogers et al. DOI:10.1002/jbt.20383]. A separate scoping review of human twin studies noted that increased DNA methylation in the GLI2 gene was associated with birth weight in singleton epigenome-wide association study meta-analyses, though this specific biomarker was not directly studied in the included twin research on birth weight discordance [Dany Laure Wadji et al. DOI:10.1371/journal.pone.0315549]. A study in humans demonstrated that Tamm-Horsfall protein (THP) is a highly specific biomarker for the forensic identification of urine [Akutsu et al. DOI:10.1111/J.1556-4029.2010.01325.X].
