This gene (IFNGR1) encodes the ligand-binding chain (alpha) of the gamma interferon receptor. Human interferon-gamma receptor is a heterodimer of IFNGR1 and IFNGR2. A genetic variation in IFNGR1 is associated with susceptibility to Helicobacter pylori infection. In addition, defects in IFNGR1 are a cause of mendelian susceptibility to mycobacterial disease, also known as familial disseminated atypical mycobacterial infection. A study in humans demonstrated that the IFNGR1 was identified as a top 10 hub gene in a sepsis-associated immune module through Weighted Gene Co-Expression Network Analysis and protein-protein interaction analysis [Jiang et al. DOI:10.1038/s41435-024-00293-4]. In a mouse model of traumatic brain injury, spatial transcriptomics revealed that the IFNGR1 was uniquely upregulated in the corpus callosum-external capsule at 7 days post-injury [Kounelis-Wuillaume et al. DOI:10.1177/08977151251390528]. A study in rats demonstrated that polytrauma (osteotomy, chest trauma, and burn) significantly upregulated local mRNA expression of the IFNGR1 at the fracture site 24 hours post-trauma compared to osteotomy alone, indicating an altered early local immune response associated with delayed bone healing and systemic inflammation [Mangum et al.
