Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. In some tissues its expression is hormonally regulated. The expression pattern of a similar mouse protein in murine developing teeth supports a role for the protein in the degradation of enamel proteins. A study in humans developed a massively parallel sequencing assay targeting coding region SNPs in body fluid-specific mRNAs to deconvolute mixtures, successfully identifying the origin and donor of blood, saliva, semen, vaginal secretion, and menstrual blood components in laboratory-generated and mock case mixtures [Liu et al. A study in humans demonstrated that the KLK4 mRNA is a highly specific marker for semen identification using Real-Time PCR assays, showing no cross-reactivity from blood, vaginal secretion, saliva, or female urine [Nussbaumer et al. The analysis of cell-specific mRNA expression, including the KLK4, represents a molecular technique for body fluid identification that supplements DNA analysis in forensic cases [Bauer DOI:10.1016/j.fsigen.2006.11.002].
