The protein encoded by this gene is a transcriptional regulator that binds as a homodimer to activating transcription factor (ATF) sites in many cellular and viral promoters. The encoded protein represses PER1 and PER2 expression and therefore plays a role in the regulation of circadian rhythm. A study in mice demonstrated that the NFIL3 is part of a common gene signature for anterior hemibody irradiation across 0.5, 2, and 10 Gy doses, with peripheral blood expression profiles predicting radiation status versus non-irradiated controls with 79-100% accuracy [Meadows et al. DOI:10.1371/journal.pone.0011535]. In human prostate tissue, the NFIL3 was identified as an upregulated mRNA marker in high postmortem interval samples, functioning as a transcription factor indicative of active postmortem gene transcription [Javan et al. DOI:10.1038/s41598-025-29561-7].
