This gene encodes a member of the steroid-thyroid hormone-retinoid receptor superfamily. Expression is induced by phytohemagglutinin in human lymphocytes and by serum stimulation of arrested fibroblasts. The encoded protein acts as a nuclear transcription factor. Translocation of the protein from the nucleus to mitochondria induces apoptosis. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2011]
A study in humans demonstrated that the NR4A1 is downregulated in the dorsolateral prefrontal cortex of subjects with Opioid Use Disorder (OUD) and is involved in regulating vascular endothelial cell proliferation and angiogenesis [Mendez et al. DOI:10.1038/s41380-021-01259-y]. Another study in humans identified the NR4A1 as a pivotal, differentially expressed mRNA in the peripheral blood nuclear cells of patients with coronary heart disease, where its expression was validated and modulated by specific lncRNAs and a miRNA in endothelial cell experiments [Liao et al. DOI:10.1186/s12920-019-0570-z]. A study in rats demonstrated that the NR4A1 mRNA expression in the iliopsoas muscle was significantly increased only by severe hypothermia, identifying it as a potential diagnostic marker for fatal hypothermia [Umehara et al. DOI:10.1007/s00414-018-1888-3]. Separately, a network analysis of human postmortem dorsolateral prefrontal cortex identified the NR4A1 as a top hub gene associated with depressed suicide, with its expression progressively decreasing from controls to MDD non-suicides to MDD suicides [Zeng et al.
