This gene encodes a member of the steroid-thyroid hormone-retinoid receptor superfamily. The encoded protein may act as a transcriptional activator. The protein can efficiently bind the NGFI-B Response Element (NBRE). Three different versions of extraskeletal myxoid chondrosarcomas (EMCs) are the result of reciprocal translocations between this gene and other genes. The translocation breakpoints are associated with Nuclear Receptor Subfamily 4, Group A, Member 3 (on chromosome 9) and either Ewing Sarcome Breakpoint Region 1 (on chromosome 22), RNA Polymerase II, TATA Box-Binding Protein-Associated Factor, 68-KD (on chromosome 17), or Transcription factor 12 (on chromosome 15). Forensic Context
A study in Wistar rats demonstrated that alcohol exposure and withdrawal significantly altered the expression of numerous mRNAs and miRNAs in the prefrontal cortex, hippocampus, and corpus striatum, with the NR4A3 being part of a downregulated gene network in the medial prefrontal cortex of post-dependent rats [Heilig et al. DOI:10.1111/Gbb.12344]. A study in humans identified the NR4A3 as a top hub gene within a co-expression network module whose progressive downregulation in the dorsolateral prefrontal cortex was significantly correlated with major depressive disorder and suicide [Zeng et al. DOI:10.1016/j.psychres.2020.113513].
