The protein encoded by this gene belongs to the protein phosphatase 1 (PP1) inhibitor family. This protein is an inhibitor of smooth muscle myosin phosphatase, and has higher inhibitory activity when phosphorylated. Inhibition of myosin phosphatase leads to increased myosin phosphorylation and enhanced smooth muscle contraction. A study in humans identified PPP1R14A as a specific gene marker defining the APOC1+/PPP1R14A+ fibroblast subcluster (FB2) within the corpus cavernosum, with its expression validated through single-cell RNA sequencing and spatial localization analysis [Zhao et al. A study in mice demonstrated that the PPP1R14A was commonly up-regulated more than 2-fold in liver tissue following in vivo irradiation of Kupffer and endothelial cells with α-particles, as validated by real-time PCR [Roudkenar et al.
