Summary

This locus encodes a member of the spectrin gene family. Spectrin proteins, along with ankyrin, play a role in cell membrane organization and stability. The protein encoded by this locus functions in stability of erythrocyte membranes, and mutations in this gene have been associated with spherocytosis type 2, hereditary elliptocytosis, and neonatal hemolytic anemia. Forensic Context

A collaborative exercise across 16 laboratories demonstrated that the SPTB mRNA marker, used for human blood identification, showed moderate sensitivity and was successfully detected by 10 out of 16 labs in fresh human bloodstains, though it was not detected in cow blood and was less sensitive than HBB in dilution series [Haas et al. DOI:10.1016/j.fsigen.2010.01.003]. Subsequent research on aged human bloodstains found that the SPTB marker, tested with both 91bp and 247bp primer sets, produced detectable signals in 50-year-old samples, with the shorter 91bp amplicon yielding more positive results than the 247bp primer, though its signals were not as strong as those from the more stable HBA and HBB markers [Zhao et al. DOI:10.1016/j.forsciint.2017.01.006]. A study in human and various animal species demonstrated that the SPTB mRNA, an erythrocytic marker, was detected in all human blood samples and some semen samples, though with significantly higher delta Ct values [Matsumura et al. DOI:10.1111/1556-4029.13045]. In a separate human study using MALDI-TOF MS for cDNA profiling, the SPTB marker for venous blood identification showed 52.4% and 95.2% positive extension reactions for its primer sets at a 20 pg cDNA input and was absent in saliva and semen samples [Donfack et al. DOI:10.1016/j.fsigen.2014.12.008].
