The protein encoded by this gene contains an HMG-box, indicating that it may be involved in bending and unwinding of DNA and alteration of chromatin structure. The C-terminus of the encoded protein is glutamine-rich due to CAG repeats in the coding sequence. A minor allele of this gene has been implicated in an elevated risk of breast cancer. A study in rhesus macaques demonstrated that whole-thorax irradiation induced significant transcriptomic dysregulation in lung tissue, where TOX3 was identified as one of the top downregulated transcripts among common differentially expressed genes in irradiated animals [Thakur et al. DOI:10.1016/j.ijrobp.2021.03.058]. This downregulation was associated with increased epithelial-mesenchymal transition, a key process in radiation-induced lung injury progression.
