The protein encoded by this gene is an isozyme of the long-chain fatty-acid-coenzyme A ligase family. Although differing in substrate specificity, subcellular localization, and tissue distribution, all isozymes of this family convert free long-chain fatty acids into fatty acyl-CoA esters, and thereby play a key role in lipid biosynthesis and fatty acid degradation. A study in humans identified the ACSL1 as a core ferroptosis-related diagnostic biomarker for heat stroke, with single-cell RNA sequencing showing its high expression in neutrophils, dendritic cells, and monocytes, and receiver operating characteristic analysis in a validation cohort demonstrating an area under the curve of 1.0 [Yin et al. Another study in humans identified the ACSL1 as a mitochondria-related hub gene significantly upregulated in acute myocardial infarction, with real-time quantitative polymerase chain reaction validation confirming its diagnostic potential, showing an area under the curve of 0.878 [Xu et al. A study in mice demonstrated that the ACSL1 transcript is part of a seven-gene set in brain tissue used for postmortem interval (PMI) prediction via linear regression, where this specific set achieved an R² of 0.95 and a slope of 0.87 between predicted and actual PMI [Hunter et al.
